Background
"The 90 kDa molecular chaperone family includes 90kDa heat shock protein Hsp90 and 94kDa glucose-regulated protein grp94, both major molecular chaperones of the cytosol and the endoplasmic reticulum. Mammalian cells express inducible Hsp90α and constitutive Hsp90β isoforms that are highly homologous and encoded by separate genes. Hsp90 chaperones a host of client proteins, stabilizing many key signaling molecules including steroid receptors, kinases, and p53 under both normal and non-physiological conditions. As many of these client proteins are known oncogenes, Hsp90 inhibitors such as the geldanamycin analog 17-AAG are known to exhibit anti-cancer activity by interfering with Hsp90¿s ATP-dependent cycle of client protein binding, leading to client protein degradation. In addition to nucleotide binding, other posttranslational modifications including acetylation and phosphorylation may play important roles in Hsp90 function. Histone deacetylase (HDAC) inhibitors have been shown to induce Hsp90 acetylation and inhibit its chaperone function. Lysine residue 294 (K294), a conserved residue in eukaryotic Hsp90, has been identified as a specific target of acetylation, inhibiting Hsp90 interaction with client proteins and kinase complex assembly in both yeast and human cells. This antibody detects Hsp90 independent of the acetylation status of the K294 residue.
