Assay Designs' hROS Detection Reagent, Aminophenyl fluorescein (APF), is a selective indicator for highly reactive oxygen species (hROS). APF is a cell permeable dye that can be used to detect hydroxyradical, peroxynitrite, and hypochlorite production in cells. This novel fluorescent reagent has little reactivity towards other ROS such as singlet oxygen, superoxide, hydrogen peroxide, nitric oxide, and alkyl peroxide. APF itself is not very fluorescent, however when reacted with hROS strong dose dependent fluorescence is exhibited. This versatile detection reagent can be adapted for use in quick one-step homogenous high throughput fluorescent plate-based assays, immunofluorescent microscopy, or flow cytometry.

Significance and Uses
Oxidative Stress describes the cellular state of oxidative damage caused by the buildup of reactive oxygen species (ROS) such as superoxides and hydroxyl radicals. Enzymatic neutralization of ROS by superoxide dismutases, catalases, and glutathione reductases maintains oxidative balance within tissues and organs. Failure of the cell to clear ROS, or to repair oxidative damage to DNA, proteins, and lipids, is commonly associated with pathogenic signal transduction, cancer, inflammation, and aging. Certain ROS species, including hydroxyradical, peroxynitrite, and hypochlorite are considered highly reactive (hROS), as they possess strong oxidizing power sufficient to directly hydroxylate aromatic rings. hROS can be selectively detected in fluorescence-based cell assays, eliminating interference from other ROS including oxygen radicals, superoxide, hydrogen peroxide, nitric oxide, alkylperoxyl radicals, or autoxidation.
