Flow cytometry is a common technique used to count, characterize, and often sort cells based on a variety of criteria including size, intracellular complexity, and fluorescence following pre-treatment of the cells with fluorescent dyes or fluorochrome-conjugated antibodies against specific antigens. Flow cytometry can be used to differentiate between cell types, particularly cells of lymphoid origin, using antibodies against lineage-specific cell surface markers. Permeabilization of cell membranes prior to antibody staining allows investigators to assess the expression of intracellular proteins as well.
