Background
Phosphatidylinositol 3-kinase (PI3-K) is a lipid kinase that phosphorylates the inositol ring of phosphatidylinositol and related compounds at the 3¿ position. The products of these reactions are thought to serve as second messengers in signaling pathways including cell growth, cytoskeletal remodeling and the trafficking of intracellular organelles. There are four subgroups (Class IA, IB, II, III) of PI3-K with distinct structure and substrate specificity. Class IA PI3-K, which functions downstream of activated tyrosine kinases, exists as heterodimers composed of a p110 catalytic subunit (alpha, beta, or gamma) and a tightly associated regulatory subunit (p85alpha, p55alpha, p50alpha, p85beta, or p55gamma). p85alpha is the major regulatory subunit of class IA PI3-Ks in most types of cells, including immune cells.
