Background
Insulin-like growth factor-I (IGF-I) is involved in regulation of neuronal growth and development in the central and peripheral nervous system. It is known to protect neurons against cell death induced by amyloidogenic derivatives, glucose or serum deprivatoin through pathways involving Akt kinase transcription factor FKHRL1 phosphorylation. Activation of the IGF system has emerged as a key factor for tumor progression and resistance to apoptosis in many cancers.

Significance and Uses
Insulin-like growth factor-I (IGF-I) was originally known as Somatomedin C but was later changed to its current name due to its structural similarity to proinsulin. IGF-I has insulin-like effects, but it appears to be involved in many other functions and has been linked to aging, nutrition, physical activity, and cancer. While each of these is very interesting on its own, together they may shed light on the role that lifestyle plays in influencing cancer risk. High levels of circulating IGF-I and low levels of IGF binding protein-3 (IGFBP-3) are associated with increased risk of several cancers. IGF-I can stimulate cell proliferation and inhibit apoptosis in many different kinds of cells. IGF-I circulates in blood bound with IGFBPs. Six IGFBPs have been identified but the majority of circulating IGF-I is complexed with IGFBP-3. The IGF-I and IGFBP-3 exist in a complex with a third protein, named the acid-labile subunit.
