Background
The Rho, Rac, and cdc42 subfamilies belong to the Rho-family of small GTP-binding proteins (G proteins or GTPases). Rho proteins have been shown to regulate actin stress fiber formation, focal adhesion and cell migration. Rho has also been shown to stimulate gene transcription and participate in Ras-induced cell transformation. Increased expression of some Rho family members appears to be a frequent event in different types of human tumors, making them potential targets for therapeutic intervention. Like all small GTPases, Rho acts as a switch regulating molecular events by cycling between an inactive GDP-bound state and an active GTP-bound state.
