Background
AIB1 (SRC3, NCoA3, pCIP, TRAM-1, RAC3, ACTR) is a member of the p160/steroid receptor coactivator (SRC) family, named for its common amplification in breast cancer. Members of this family include a defined nuclear receptor box motif (LxxLL) that is critical for binding to nuclear hormone receptors, and two C-terminal activation domains that bind secondary coactivators such as the p300/CREB binding protein (CBP) and CARM-1 to confer acetyltransferase or methyltransferase downstream signals, respectively. Phosphorylation of AIB1 by IkB kinase (IKK) has been shown to regulate translocation of AIB1 from the cytosol to the nucleus. AIB1 binds to estrogen receptor (ER), and its overexpression in hormone responsive cancers, and frequent amplification in breast and other cancers make it a focus for the treatment of cancer and the study of hormone receptor signaling.
