Background
Melanocortin receptors serve as the principle receptors for the pro-opiomelanocortin (POMC) family of peptide agonists, as well as agouti and agouti-related protein (AGRP) inverse agonists. POMC derived peptides include adrenocorticotrophin (ACTH), melanocyte-stimulating hormones (alpha, beta and gamma-MSH), and beta-endorphin. Five known melanocortin receptors (MC1-MC5) display distinct and relatively localized expression patterns that contribute to their physiological activity. The MC1 receptor is expressed primarily by melanocytes and regulates pigment production, while the MC2 receptor coordinates ACTH-stimulated glucocorticoid production in the adrenal cortex. The neuronally expressed MC3 and MC4 receptors bind MSH peptides and serve as primary receptors for the antagonist AGRP, signaling primarily via Gs alpha subunits to stimulate cAMP production. MC4 receptors are implicated in energy balance disorders leading to obesity, and are known to play a role in erectile function.
