Background
Progression through the cell cycle is controlled by cyclin-dependent kinases (CDKs). Human p21WAF1/CIP1 is a CDK inhibitor that was first cloned and characterized as a mediator of p53-induced growth arrest, and serves as a regulator of cell cycle progression, terminal differentiation, and apoptosis. Human p21 inhibits cyclin-CDK2 and cyclin-CDK4 complexes, regulating cell cycle progression in G1 phase. p21 expression can be induced independently of p53 by TGF-beta, tamoxifen, progesterone, nerve growth factor and apicidin. Human p21 expression has been implicated in a variety of human cancers including those of the prostate, bladder, and esophagus.
