Methotrexate is an antimetabolite and antifolate drug that acts by inhibiting the metabolism of folic acid. In higher doses (5000 mg/week) it is effective as an anti-cancer agent, while in lower doses (5-25 mg/week) it is a first-line drug in the treatment of rheumatoid arthritis non-responsive to nonsteroidal anti-inflammatory drugs.
