Background
Nitric oxide (NO) is synthesized from L-arginine, oxygen, and NADPH by three known isoforms of heme-containing flavoproteins termed NO synthases (NOS I-III). Analysis of cDNA clones has identified three distinct NOS genes in mammals: neuronal (nNOS/NOS-I), endothelial (eNOS/NOS-III), and inducible (iNOS/NOS-II). eNOS and nNOS are constitutively expressed, agonist-triggered, Ca2+/calmodulin-dependent, and are inhibited by L-arginine analogues (L-NNA, L-NMMA). They are expressed in endothelium, adrenal glands, brain, and platelets. iNOS is inducible, Ca2+/calmodulin-independent, and also inhibited by NMMA and L-NNA. It is expressed in macrophages, hepatocytes, tumor cells, vascular smooth muscle and endothelial cells.
