Background
The Insulin receptor substrate family, which consists of IRS 1-4, consists of cytoplasmic docking proteins involved in the regulation of metabolism and proliferation by Insulin, IL-4, and other cytokines. Upon binding of insulin to its receptor, the receptor is activated leading to the cross-phosphorylation of tyrosine residues within the cytoplasmic domain. These P-Tyr motifs serve to recruit a variety of Src homology-2 (SH2) domain-containing proteins such as PI3-kinase, Grb-2, SHP-2, Nck and Crk.
